چکیده
The epidermis is a spatially arranged tissue of different keratinocytes consisting stem cells, proliferating and differentiating cells. However, the mechanisms regulators arrangement remain unknown. Here, we show that serum starvation induces spontaneous pattern formation in keratinocytes. When were cultured beyond confluence, areas with densely clustered cells those sparsely distributed appeared alternately. This was triggered by but lifted supplementation fresh serum. Gene knockout experiments, inhibitor treatment, mathematical modeling revealed cell-cell adhesion through adherens junctions essential sufficient for formation. poised to differentiation, while prone proliferate. YAP signaling pathway modulated this switching between differentiation proliferation states sensing junctions. Finally, three-dimensional epidermal cultures, serum-deprived condition formed thicker than serum-rich condition. These data demonstrate starvation-induced underlies keratinocyte patterning helps stratification epidermis.

Calcium-induced assembly of adherens junctions in keratinocytes
Extracellular calcium concentration has been shown to control the stratification of cultured keratinocytes, presumably by regulation of formation of desmosomes. Previous studies have shown that keratinocytes cultured in medium containing 0.1 mM Ca++ form loose colonies without desmosomes. If the Ca++ is raised to 1 mM, desmosomes are assembled and the distribution of keratin filaments is altere...

How did genetic analysis get started in this system? Here we depended on a breakthrough from the lab of Peter Wolk and Jeff Elhai at Michigan State. They showed that DNA could be transferred by conjugation from E. coli to Anabaena. Postdoc Bill Buikema first isolated mutants incapable of fixing nitrogen, most of which could not differentiate heterocysts. He then used a cosmid library of wild-ty...

James Nelson (Stanford University School of Medicine, Stanford, CA). There is no direct link, they say, between cadherin, via and -catenin, to actin. Weis’s original goal was ambitious—a structu...
متن کاملFormin' adherens junctions
pithelial cells zipper together thanks to linear actin cables—cables that assemble at newly formed adherens junctions to stabilize them, thereby counteracting retractive forces at sites of cell–cell contact. Now, Agnieszka Kobielak, Amalia Pasolli, and Elaine Fuchs (Rockefeller University, New York, NY) have found that formin-1 drives the actin polymerization that creates the cables. Formin-1 e...
متن کاملRAC1 regulates adherens junctions through endocytosis of E-cadherin.
The establishment of cadherin-dependent cell-cell contacts in human epidermal keratinocytes are known to be regulated by the Rac1 small GTP-binding protein, although the mechanisms by which Rac1 participates in the assembly or disruption of cell-cell adhesion are not well understood. In this study we utilized green fluorescent protein (GFP)-tagged Rac1 expression vectors to examine the subcellu...
